氟化钠致小鼠染色体畸变和精子畸形

The Effects of Sodium Fluoride on Chromosome Aberration of Bone Marrow and Sperm Malformation in Mice

摘要 用氟化钠诱发小鼠骨髓细胞染色体畸变和生殖细胞精子畸形,同时用氯化钙进行抗诱变的观察。结果一、用氟化钠进行诱发,当剂量为1/2 LD50mg kg 时,染色体畸变率为26.0%;1/5时为19.6%;1/10时为20.0%;1/20时为12.4(?):环磷酰胺阳性组畸变率为33.5%;阴性对照组畸变率为0.01%。精子畸形发生率1/5时为4.63%;1/10时为5.03%;1/20时为3.72%;阴性对照组为2.38%,均有显著性差异。提示氟化钠在高剂量下可诱发中度或低度的染色体畸变和精子畸形。二、用氯化钙抗诱变时,当剂量为1/5 NaF+2mg/g CaCl_2时,染色体畸变率有所下降,但效果尚不显著,有待寻找更有效的抗诱变剂(Antimmtagen) The result of the frequency of chromosome aberration in mice induced by the sodium fluoride and calcium chloride antiaberration was reported.There were four experiment groups S.fluoride in doses 50mg/kg(1/2 LD_(50)).20mg/kg (1/5 LD_(50)).10mg/kg(1/10LD_(50)).5 mg/kg(1/20 LD50),and were three experiment groups given S.fluorideand calcium chloride in doses NaF 20mg/kg, +CaCl_2 2mg/g.NaF 10mg/kg+CaCl_2 1mg/g,and NaF 5mg/kg+CaCl_2 1mg/g respectively.Cyclophosphamide 50mg/kg was used as positive control and normal saline solution as negative control.In all positive control and experiment groups,typical positive mutation changes were observed.The chromosome abnormality rate on the bone marrow cells:S.fluoride(1/2 LD_(50)) 26.0%,S.fluoride(1/5LD_(50))19.6%,S.fluoride(1/10LD_(50))20.0%,S.fluoride (1/20LD_(50) 42.4%and S.fluoride 1/5LD_(50)+2mg/gm C.chloride 14.4%;S. fluoride 1/10LD50+1mg/gm C.chloride 12.0%;S.fluoride 1 /20LD50+1mg/g C.chloride 9.0%.Cyclophosphamide 33.5%.In negative control groups,a all was negative(0.01%).We compared the two experimental groups and discussed them.Depending upon the results,author think that some genetic effect are observed over the dose 5mg/kg of S.fluoride.The calcium chloride was a medicament of antiaberration.It is reported otherwise in this article that the mixture of S.fluoride was administered to three groups of male mice by injection for five consecutive days with doses of 20mg/kg,10mg/kg and 5mg/kg respectively.There were significant change in spermmorphology.The result showed that the mixture absorbed by the mice could affect the normal development of the mouse's sperm of induce the sperm malformation.
