酮对大鼠心肌线粒体氧化损伤的保护作用 被引量:9

摘要 目的 研究 3种穿心草 口山酮 (xanthone ,Xan)对心肌线粒体氧化损伤的保护作用。方法 采用抗坏血酸和硫酸亚铁为线粒体损伤诱导剂 ,探讨Xan对线粒体的膨胀、脂质过氧化及膜流动性的影响。结果  3种Xan不同程度地减少受伤线粒体的膨胀作用及阻止脂质过氧化的形成 ,增加膜脂质流动性 ,维护线粒体结构和功能的完整 ,作用强度依次为Xan Ⅰ >Xan Ⅲ >Xan Ⅱ。结论  3种Xan对心肌线粒体氧化损伤均有不同程度的保护作用 ,作用机制可能与其清除自由基、抗脂质过氧化作用有关 ,而 OBJECTIVE To study the protective effects of xanthones:1,8 dihydroxy 3,5 dimethoxyxanthone (Xan Ⅰ),1 hydroxy 3,5 dimethoxy xanthone (Xan Ⅱ) and 1 hydroxy 3,7,8 rithoxyxanthone (Xan Ⅲ),isolated from Canscora lucidissima on injured rat heart mitochondria and to investigate its protective mechanisms.METHODS The injured rat heart mitochondria was induced by ascorbate and FeSO 4,with biochemical methods.Some indicates related to lipid peroxidation of mitochondria were measured.RESULTS It was found that 0.02 μmol·L -1 of three Xans increased the membrane fluidity of the injured mitochondria,decreased the lipid peroxidation and mitochondria swelling.The intensity of above effects among Xans was Xan Ⅰ>Xan Ⅲ>Xan Ⅱ.CONCLUSION Three components of Xans protected the rat heart mitochondria from injury.It may be associated with the function of scanging free radical and antioxidation of xanthones.The results suggested that the amount and place of OCH 3 and OH may be associated with the activity of xanthones.
