摘要
目的 研究抗苗勒管激素 (anti mullerianhormone ,AMH)在诊断卵巢颗粒细胞瘤(granulosacelltumor,GCT)中作为特异性肿瘤标记物的实验依据。方法 采用免疫组织化学、原位杂交、Northern杂交法研究转基因小鼠早、晚期卵巢GCT中AMH及其受体mRNA的表达 ,并用放射性碘( 12 5I)标记法、Northern杂交法研究经培养的卵巢GCT细胞中AMH及其受体mRNA的表达。结果 转基因小鼠早期卵巢GCT中 ,AMH的免疫组织化学检测和AMH受体mRNA的原位杂交检测结果显示 ,两者的阳性表达率均为 10 0 % ;而晚期卵巢GCT中 ,两者的阳性表达均为 8例中 2例 ,分别比较 ,差异均有极显著性 (P <0 .0 1)。Northern杂交研究同样显示 ,早期卵巢GCT和卵巢GCT细胞AMH受体mRNA的阳性表达率均为 10 0 %。放射性自显影的研究显示 ,在卵巢GCT细胞膜处有放射性亮点 ,12 5I标记的AMH与卵巢GCT细胞结合率达 10 0 %。结论 AMH可作为一种特异性的肿瘤标记物 ;可用于卵巢疑难肿瘤的鉴别诊断以及早期卵巢GCT的诊断 ;
Objective Toprobetheexperimentalbasisforanti mullerianhormone(AMH)servedasa specifictumorlabelindiagnosisofgranulosacelltumor(GCT)ofovary .Methods TheexpressionofAMHandits receptormRNAinearlyandlateGCTofovaryintransgenicmicewasstudiedbyusingimmunohistochemistry,in stiuhybridizationandNorthernhybridization .Furthermore ,theexpressionofAMHanditsreceptormRNAin culturedGCTcellswasalsodeterminedbyusingradioiodinelabelingmethodandNorthernhybridization .Results IntheearlyovaryGCToftransgenicmice ,immunohistochemistryshowedthatAMHwaspositivelystainedas brownishyellowgranulars,whichwerelocatedinthecellmembraneofthetumor,insituhybridizationshowedits receptormRNAwaspositiveasbluegranulars ,whichwerealsolocatedinthecellmembrane.Thepositiverateof bothAMHanditsreceptormRNAwas 10 0 % .Resultsbynorthernhybridizationalsoshowedthatthepositiverate ofAMHanditsreceptormRNAintheearlyovaryGCTandGCTcellswas 10 0 % .Radioautographyindicatedthat therewasaradioactivebrightpoint,thebindingrateof 12 5IlabeledAMHwithtumorcellsreached 10 0 % . Conclusions AMHcanbeservedasaspecifictumormarkerinthediagnosisofdifficultandcomplicatedtumors ofovary,andinthediagnosis,treatmentandfollow upsurveyoftheearlyGCT .
