甜樱桃杂交后代自交亲和性鉴定 被引量:4

摘要 利用SFB4'基因特异引物BFP200/BFP201与内对照引物PCR IC-F/IC-R对甜樱桃自交亲和品种斯特拉自然杂交后代的29个株系的基因组DNA进行扩增,鉴别其是否具有自交亲和性,并对已进入结果期的10个株系进行田间自花授粉试验进一步验证其自交亲和性。结果表明:斯特拉杂交后代的29个株系中27.6%的株系检测出自交亲和特异条带,具有自交亲和性。在自交亲和基因鉴定时出现特异条带的品种和株系,其自交结实率均≥18.6%;而未检测出自交亲和基因特异条带的品种和株系,其自交结实率均≤14.8%。甜樱桃自交授粉结实率>15.0%的品种,即可认为具有自交亲和性。 The genomic DNA of 29 natural hybrid progenies of sweet cherry Stella was amplified using the specific primers BFP200/BFP201 of SFB4' gene and the control primers IC-F/IC-R to identify the self-compatibility of hybrid progenies.The self-compatibility of 10 progenies was verified through self-pollination experiment.The results showed that 27.6% natural hybrid progenies had the specific band,which were self-compatible.The self-fruitful rate of varieties and progenies with specific band and those without specific band was greater than or equal to 18.6% and lower or equal to 14.8%,respectively.The varieties and progenies were self-compatible when the self-fruitful rate was greater than 15.0%.
