微生物产生的免疫抑制活性化合物S-9818的研究 Ⅱ.发酵及生物学性质

摘要 千佛链霉菌新种 SIIA9818(Streptomycesqianf oensis sp nov SIIA9818)在含合适碳氮源的培养基内 ,经发酵获约 50 %菌丝量的培养液。发酵第 5天 ,其代谢产物 S-9818的产生达到高峰 ,主要存在于菌丝体内。S-9818对革兰氏阳性、阴性菌和酵母不显示抗微生物活性 ,但有显著的抗其它真菌性质 ,特别是对黑曲霉、烟曲霉有极强的抗菌作用。淋巴细胞产生抑制试验显示 ,S-9818比环孢素有更强的抑制小鼠淋巴细胞产生的免疫抑制性能 ,S-9818的生物学性质与日本报道的 FK-50 6十分相似。 In a common medium with suitable C, N sources, Streptomyces qianfoensis sp. nov. SIIA 9818 was cultured for 7 days, harvested cultural broth with 50% mycelial mass. A metabolite named S 9818 was obtained in a peak amount at 5th day in the mycelia mainly. S 9818 appeared no activities against G +, G - bacteria, neither yeast, but active against the fungi, Aspergillus niger and A.fumigatus , especially. In the inhibition tests of lymphocyte proliferation in mice, S 9818 demonstrated its immuno inhibiting properties, it has more stronger effects of inhibiting lymphocyte proliferation than those of Cyclosporin A. As for its biological properties, we found that S 9818 is very similar to FK 506, an immunosuppressant developed in Japan.
