直接免疫荧光法与多重逆转录聚合酶链反应对呼吸道感染常见病毒的诊断价值 被引量:3

Diagnostic value of direct immunofluorescence and multiplex reverse transcription polymerase chain reaction for common viral respiratory tract infections

摘要 目的探讨直接免疫荧光法(DFA)和多重逆转录聚合酶链反应(RT-PCR)对呼吸道常见病毒人鼻病毒(HRV)感染患者分泌物的检测,判断其临床诊断价值。方法选取2010年2月-2011年12月收集的急性呼吸道感染患儿鼻咽分泌物210例,分别用DFA和RT-PCR进行检测,对两组测定方法的检测结果进行判定。结果多重RT-PCR检出112份阳性标本,阳性率为53.33%,DFA检测出89份阳性标本,阳性率为42.38%,两组比较,差异有统计学意义(χ2=27.623,P<0.01)。结论 DFA检测方法可以广泛适用于临床检测中;多重RT-PCR检测方法的敏感性高,检测病毒的范围广,可以做亚型鉴定方法,适用于呼吸道病毒流行病学的调查研究。 OBJECTIVE To investigate the detection of direct immunofluorescence (DFA) and multiplex reverse transcription polymerase chain reaction (RT-PCR) on the secretions of patients with common respiratory virus human rhinovirus (HRV) infection, and to determine its clinical diagnostic value. METHODS A total of 210 cases of nasopharyngeal secretions obtained from the children with acute respiratory tract infections from Feb 2010 to Dec 2011 were chosen, and they were respectively detected by DFA and RT--PCR for determination, and the test results of the two test methods were determined. RESULTS The positive rate was 42.38% by the DFA detection (89/210), and the positive rate was 53.33% by multiple RT-PCR (112/210), the difference was statistically significant (X^2 = 27. 623, P〈0.01). CONCLUSION DFA detection can be widely used in clinical testing. With the high sensitivity and wide range of detection of the virus subtype identification, the multiplex RT-PCR is suitable for the research of respiratory virus epidemiology.
