麻疯树FAD3基因的克隆及亚细胞定位研究 被引量:3

摘要 为研究麻疯树JcFAD3的亚细胞定位,本实验从麻疯树叶片中克隆了JcFAD3基因,将其与绿色荧光蛋白基因融合构建植物表达载体。利用基因枪转化的方法将重组载体转入到洋葱表皮细胞中进行瞬时表达,通过检测融合蛋白在洋葱内表皮细胞中的分布来对JcFAD3基因进行亚细胞定位,荧光显微镜检测结果表明,JcFAD3基因表达产物定位于细胞质中。 To identify the subcellular location of JcFAD3 of Jatropha Curcas L., JcFAD3 was cloned form Jatropha leaves and fused in-frame to a green fluorescent protein gene in order to construct the recombinant vector pCMBIA1304-FAD3. The recombinant vector pCMBIA1304-Fad3 was transformed into onion epidermis cells by the gene gun method and the transient expression of JcFAD3 and GFP fusion protein in onion epidermal cells showed that JcFAD3 protein was localized in the cytoplasm.
