生物信息资源在耻垢分枝杆菌EmbB多克隆抗体制备中的应用

摘要 目的建立通过生物信息学手段制备耻垢分枝杆菌(M.smegamatis)EmbB(Sm EmbB)多克隆抗体的方法。方法应用生物信息资源在Sm EmbB的N端寻找一段18个氨基酸的特异短肽,合成后将其与白喉类毒素分子相连,以所形成的复合物免疫动物制备抗Sm EmbB的多克隆抗体。结果 Western blot结果显示获得的抗体可特异地作用于Sm EmbB。结论通过生物信息学手段制备多克隆抗体方法的建立解决了对蛋白质功能研究中缺乏抗体的困扰,为特殊蛋白质的抗体制备提供了借鉴。 Purpose To build up a new method to make polycolonal antibody of EmbB in M. smegma- tis by utilizing bioinformation resource. Methods A peptide of Sm EmbB with 18 amino acids was de- signed based on to the analysis of bioinformation, and linked with Diphtheria Toxoid to form a complex which was utilized to immune mouse for getting Sm EmbB's polycolonal antibody. Results The antibody achieved can work on Sm EmbB well with being identified by Western blot. Conclusion This method can provide a new point of view to obtain polycolonal antibody for a certain protein without antibody available.
