摘要
目的 探讨碱性成纤维细胞生长因子 (bFGF)对离体培养的生后大鼠前脑脑室下层 (SVZ)神经元前体细胞增殖、分化的影响。方法 取体重 10 0~ 15 0g雄性SD大鼠SVZ细胞进行离体培养 ,培养液分别为NeurobasalMedium/B2 7和NeurobasalMedium/B2 7+bFGF。倒置显微镜下观察其生长变化的形态学特征 ,并作细胞计数 ;免疫细胞化学染色观察其神经化学性质。结果 ①培养液中添加bFGF者 ,其SVZ源神经细胞的数目明显多于未添加bFGF者 ;②在该 2种培养液条件下 ,SVZ源神经元前体细胞都能分化为成熟神经元 ,且其中的大多数神经元是γ-氨基丁酸能神经元 ,只有少量神经元是谷氨酸能或多巴胺能神经元。结论 ①bFGF能促进离体培养的生后大鼠SVZ源神经元前体细胞的增殖 ;②在特定的培养条件下 ,生后大鼠SVZ源神经元前体细胞能分化为不同性质的神经元。
Objective To observe the effect of the basic fibroblast growth factor (bFGF) on the proliferation and differentiation of the neuronal precursor cells derived from the subventricular zone (SVZ) of the postnatal rat forebrain in vitro. Methods The SVZ cells from SD rats of 100~150 g body weight were cultured in the Neurobasal Medium supplemented with B27 and that supplemented with B27+bFGF separately. The morphological phenotypes of the SVZ-derived neuronal cells were observed and the cell count of them was made under the IX50/IX70 biological light microscope(Olympus). And the chemical phenotypes of them were indentified by means of immunocytochemical staining. Results (1)The number of neuronal cells derived from the SVZ was greater in the Neurobasal Medium supplemented with B27+bFGF than in that suplemented with B27 alone. (2)The precursor cells derived from the SVZ became mature in both culture media. Most of the neuronal cells were GABA-positive and only a few of them were Glu-positive or DA-positive. Conclusions (1)bFGF can stimulate the proliferation of neuronal precusor cells derived from the SVZ of postnatal rat forebrain in vitro. (2)These neuronal precursor cells can differentiate into different nerons, which may be GABA-positive, Clutamate-positive or DA-positive.
