摘要
目的 探讨神经生长因子 (NGF)对癫所致脑损伤有无拮抗作用。方法 采用红藻氨酸 (KA)诱导癫发作大鼠模型 ,分别于实验 12、2 4、48h观察急性癫发作后大鼠海马CA1区的超微结构和光镜结构的改变及NGF对它们的影响。结果 实验 12h ,大鼠海马CA1区神经细胞变性坏死 ,神经胶质细胞水肿变性 ,毛细血管内皮肿胀 ;实验 2 4h上述病理改变加重 ,实验 48h最为严重。神经细胞存留数渐次下降 ,48h为最低。给予NGF脑室内注射后 ,在各相应时间点病理改变相对减轻 ,神经细胞存留数明显增加 (P <0 .0 1)。结论 NGF能抑制癫所致脑损伤 ,NGF对癫脑损伤有保护作用。
Objective To explore whether exogenous nerve growth factor (NGF) might inhibit cerebral injury owing to acute seizures or not. Methods With the model of epilepsy induced by kainic acid and NGF injected into the cerebral ventricle,the histologic and ultrastructural changes in the hippocampus CA1 were observed at the 12,24,48th hour following the experiment beginning.Results At hour 12,the degeneration and necrosis of neurons, neuroglial cells and vascular endothelial cells were found .At hour 24,the cerebral lesions as indicated above were more severe than the former group. At hour 48,histologic changes were the most severe of three groups. Neurons in CA1 of hippocampus decreased step by step. There were the fewest such neurons 48 hours after. Injected NGF into the cerebral ventricle, the rats had milder cerebral lesions and more neurons than those not injected NGF and killed at the same time (P<0.01). Conclusions NGF can inhibit the cerebral injury induced by seizures. NGF has protective effect on the cerebral injury owing to epilepsy.
