摘要
为建立冬虫夏草虫草甘露醇含量测定方法,从冬虫夏草的组织直接切取虫草菌核内部的一小块菌索,采用优化好的培养基配方来培养冬虫夏草,并从其菌丝体中提取、分离虫草甘露醇。采用比色法测定甘露醇的含量。人工发酵菌丝体中甘露醇的含量为4.23%,平均加样回收率为100.76%,变异系数(RSD)为1.92%,天然虫草子实体中甘露醇的含量为11.0%,平均加样回收率为100.52%,RSD为1.38。此法测定虫草甘露醇具有简单快速、灵敏度高、精确度和准确性高、重现性好,可作为检测冬虫夏草甘露醇含量的优选方法。
The a little piece of shoestring was obtained from the interior Cordyceps sinensis sclerotium, which derived from the organization of Cordyceps sinensis, and used the optimizing on liquid culture medium to culture the Cordyceps sinensis, from which the mannitol was extract and isolated. Trough the method of colorimetric assay the mannitol contents were determinated and the determinate way of the mannitol contents was established. The mannitol contents of manual mycelium were 4.23%, and the average recovery rate was 100.76%, with the RSD was 1.92%. The mannitol contents of wild Cordyceps fruit body was 11.0%, and the average recovery rate was 100.52%, with the RSD was 1.38%. This method was simpler, quicker, higher accuracy and veracity, and better recurrence. It could be used to be preferred method in the determination of the polysaccharide and mannitol in Cordyceps sinensis.
