红蘑多糖的纤维柱层析分离纯化 被引量:6

Separation and purification of Gomphidius rutilus polysaccharides by column chromatography

摘要 选择野生红蘑作为提取多糖的原料,采用预处理后的DEAE-52纤维素粉末制成OH-型柱对红蘑中的多糖进行分离纯化,结果表明:在上样量为200mg、洗脱速度为0.5mL/min的条件下洗脱,得到4个主峰,依次为GRPⅠ、GRPⅡ、GRPⅢ和GRPⅣ,其中GRPⅠ出现在水洗脱部分,GRPⅡ、GRPⅢ和GRPⅣ分别出现在0.1mol/LNaCl、0.3mol/LNaCl、1.0mol/LNaCl洗脱部分。经计算,GRPⅠ、GRPⅡ、GRPⅢ和GRPⅣ分别占12.6%、8.7%、22.3%和27.6%。 The primary purification of the polysaccharides extracted from Gomphidius rutilus was carried out by DEAE-52 cellulose chromatography, which was as a vector after pretreatment. The results show that, the four kinds of pure polysaccharide named GRP I, GRPII, GRPIII and GRPIV were mainly obtained when the adding sample was 200 mg and the rate of flow was 0.5 mL/min. GRPI appeared undergoing distilled water elution in column chromatography. GRPII, GRPIII and GRPIV appeared undergoing NaCI gradient elution with the concentration 0.1 mol/L, 0.3 mol/L and 1.0 mol/L in column chromatography. The ratio of them were12.6%, 8.7%, 22.3% and 27.6% respectively.
