摘要
以Ficoll-400为原料,采用氯乙酸法在碱性条件下进行羧甲基化改性。以产物取代度(DS)为评价指标,通过单因素和正交实验对改性条件进行优化。结果表明:溶胀介质为85%乙醇,n(聚蔗糖):n(氯乙酸):n(氢氧化钠)=1:4.8:5.5,反应温度为70℃,反应时间为10h时,产物的DS可达0.6412。该改性方法工艺简单,易于控制,可以为其他多糖的羧甲基化改性提供参考。
Using Ficoll-400 as the raw material, carboxy methylation was studied by adopting chloroacetic acid in alkaline conditions, through single factor and orthogonal experiments, the effects of reaction conditions no degree of substitution (DS) were investigated. The result showed that the DS product was 0.6412 when reaction medium was 85% ethanol, n(FicolI-400):n(CH2CICOOH):n(NaOH) equaled to 1:4.8:5.5 with reaction time of 10 h in 70 %. This carboxymethylation has simple process and was easy to control, which can provide reference to the carboxymethylation of other polysaccharides.
