摘要
目的探讨雌二醇(E2)、孕酮(P)对人子宫内膜癌细胞体外增殖及凋亡的影响。方法体外培养子宫内膜癌细胞,采用流式细胞仪观察不同浓度E2、P和E2+P对细胞周期及细胞凋亡率的影响、MTT比色法观察HEC-1B细胞的增殖。结果流式细胞仪分析,E2组细胞周期S期比例上升,凋亡率下降;P组和E2+P组G0/G1期上升,S期下降,细胞凋亡率上升;MTT结果显示,随着孕酮浓度的增加,E2对HEC-1B细胞有明显促生长作用(P<0.01),P和E2+P对细胞生长有明显抑制作用(P<0.01),增殖抑制率与浓度呈正相关(P<0.05)。结论 E2对HEC-1B细胞具有促进增殖作用;P则抑制HEC-1B细胞增殖,并诱导其凋亡;E2+P显示了抑制增殖、诱导凋亡的作用增强。足量的P可拮抗E2对内膜癌细胞的促增殖作用。
Objective To investigate the impact of estradiol and progesterone on proliferation and apoptosis of endometrial cancer cell line in vitro. Methods Endometrial cancer cells were cultured in vitro. The treated groups consisted of estradiol group, progesterone group and estradiol plus progesterone group. Cell cycle, apoptotic percentage were detected by flow cytometry. The proliferationat of endometrial cancer ceils was determined by methyl thiazolyl tetrazoliurm (MTT) method. Results FCM showed that estradiol increased the proportion of S phase, reduced the apoptotic rate. And the proportion of G0/G1 phase and the apoptotic rate were increased significantly in progesterone groups(single or combined), the proportion of S phase was reduced. MTT showed that with the increasing of the concentration of estradiol, they stimulated the growth of endometrial cancer cells significantly, and the progesterone group inhibited the growth of endometrial cancer cells significantly. Conclusion Eestradiol could enhanced the proliferation and inhibit its apoptosis in endometrial cancer ceils. Progesterone could inhibit the proliferation of endometrtal cancer cells and induce apoptosis. Enough progesterone can resist the role of proliferation enhancement by estradiol.
