氯化三乙基锡抑制C6胶质瘤细胞增殖及机制

摘要 目的探讨氯化三乙基锡(TETC)对C6胶质瘤细胞体外增殖抑制作用的机制。方法采用MTT法、荧光原位末端标记方法和Western印迹方法,分别检测不用浓度TETC作用C6胶质瘤细胞48 h后细胞增殖抑制率、细胞凋亡率和Bcl-2和Bax蛋白的表达水平。结果 TETC对C6胶质瘤细胞具有抑制增殖作用,增殖抑制率呈剂量依赖性;TETC可诱导C6胶质瘤细胞凋亡,细胞凋亡率呈剂量依赖性上升;TETC可下调Bcl-2和上调Bax蛋白表达水平。结论 TETC可抑制C6胶质瘤细胞体外增殖并诱导其凋亡,机制可能与其下调Bcl-2和上调Bax蛋白表达有关。 Objective To study the inhibition mechanism of triethyltin chloride (TETC) on proliferation of C6 glioma cells in vitro. Methods Inhibitory rate of TETC on the proliferation of C6 glioma cells , apoptotic rate of C6 glioma cells and expression of Bcl-2 and Bax protein were tested by MTF assay, fluorescent TUNEL assay and Western blotting analysis at 48 h after treated by different concentrations of TETC on C6 glioma cells, respectively. Results The proliferation of C6 glioma cells was inhibited by TETC in a dose-dependent manner. TETC induced apoptosis of C6 glioma cells and the apoptotic rate was increased in a dose-dependent manner. The expressions of Bcl-2 and Bax protein were down-regulated and up-regulated, respectively. Conclusions TETC could inhibit the proliferation of C6 glioma cells in vitro. TETC could induce the apoptosis of C6 glioma cells, decrease the expression of Bcl-2 protein, increase the expression of Bax protein, lead to apoptosis of C6 glioma cells, and these maybe the mechanism of anti-proliferation of TETC.
