慢性激怒应激对大鼠血和结肠粘膜前列腺素E_2、环核苷酸的影响

Effects of Chronic Irritation Stress on Prostaglandin E_2 and Cyclic Nucleoside of Colon Mucosa and Blood of Rats

目的 观察慢性激怒应激对大鼠血及结肠粘膜中前列腺素 E2 (PGE2 )、 c AMP、 c GMP含量的影响 ,探讨其作用和意义。方法 大鼠用夹子将其后肢夹住 ,直立位挂起 ,并让大鼠之间出现扭打嘶咬。钳夹时间第 1天为 30 min,以后每天增加 10 min,连续 2周。以不作任何处理的大鼠作正常对照。造模结束后取血浆及结肠粘膜 ,以放免法测定血及结肠粘膜中 PGE2 、 c AMP、c GMP含量。结果 大鼠血中 PGE2 、c AMP、c GMP含量和 c AMP/ c GMP与正常对照组之间无明显差异 ;大鼠结肠粘膜中 PGE2 、c AMP的含量及 c AMP/ c GMP较正常组明显升高 (P<0 .0 5 ,P<0 .0 1) ;而 c GMP的含量则与正常组之间无显著差异。结论 慢性激怒应激可引起大鼠结肠粘膜 PGE2 和环核苷酸的变化 ,这些变化可能在胃肠功能紊乱性疾病中起一定的作用。

To observe the changes of the PGE 2 and cyclic nucleosides contents of the plasma and the colonic mucosa in the stressed rats.Methods:The rats were hung by clipping their posterior limbs,and made to wrestle and bite each other.Thirty minutes were taken in the first day,and added ten minutes in the next day,then from this went on for two weeks,the normal group was used as control.After the models were made,the plasma and the colonic mucosa of all the rats were taken,and the contents of PGE 2,cAMP and cGMP were measured using RIA.Results:There were no significant difference of the plasma levels of PGE 2,cAMP,cGMP and cAMP/cGMP between the two groups.The levels of PGE2,cAMP and cAMP/cGMP in the colonic mucosa were significantly higher than those of normal control group,but the level of cGMP in that had no significant difference between the two groups.Conclusions:There were some changes of PGE2 and cyclic nucleosides contents of the colonic mucosa in the stressed rats,and those changes had certain effect on the diseases of the gastrointestinal functional disorders.
