人参皂甙Rh_2对C_6胶质瘤细胞的增殖抑制和诱导凋亡作用

Inhibition of growth and induction of apoptosis of Ginsinoside Rh_2 on cultured C_6 glioma cells

目的 :观察人参皂甙 Rh2 对 C6胶质瘤细胞的增殖抑制和诱导凋亡作用 ,并在分子水平探讨其作用机理。方法 :细胞计数法检测细胞增殖 ,流式细胞仪分析细胞周期和凋亡。结果 :1 Rh2 2 ,4,8μmol/L对 C6胶质瘤细胞有增殖抑制作用 ,且呈明显量效关系 ;2 Rh2 作用 72 h后 ,4μmol/L浓度对 C6胶质瘤细胞诱导凋亡作用最为明显 ;3 Rh2 作用后 ,G0 /G1期细胞百分率明显下降 ,而S期细胞百分率显著增加。结论 :Rh2 对 C6胶质瘤细胞有明显的增殖抑制和诱导凋亡作用 ,且这种作用有周期特异性。

Objective:To investigate Ginsinoside (G Rh 2′s) function of suppressing growth,inducing apoptosis of glima C 6 cells and to find the mechanism of this effect at molecular level.Methods:Cell proliferation was evaluated by cell numeration.And cell proliferation cycle and apoptosis were tested by flow cytometry (FCM).Results:①G Rh 2 exerted a dose dependently inhibitory effect on cell proliferation at the dose between 2~8 μmol/L.②After treated with G Rh 2 of 4μmol/L for 72 h,the most significant apoptosis effect on C 6 cells was induced.③Exposure to G Rh 2 resulted in obvious increase in S phase and decrease in G 0/G 1 phase of C 6 cells.Conclusion:G Rh 2 can inhihit proliferation and induce apoptosis of C 6 cells and this effect is cycle specific. 〔
