对IL-4两种测活方法的比较及MTT法最适条件确定 被引量:14

摘要 目的建立一种稳定、灵敏的方法用于检测 IL - 4生物活性。方法比较人外周血 T淋巴细胞与 CTL L - 2 /IL - 4R细胞系对 rh IL - 4的增殖应答特点及 MTT法用于 rh IL - 4生物活性检测过程中的细胞浓度、细胞培养时间、MTT掺入浓度及时间等条件。结果两者均能对 rh IL - 4呈规律性增殖应答。 IL - 4活性测定时 ,人外周血 T淋巴细胞检测最低限为 8U /ml,CTL L - 2 /IL - 4R细胞系为 0 .73U /ml。确定了 MTT法用于 CTL L - 2 /IL - 4R细胞系检测 rh IL - 4生物活性时的最佳条件方案。结论就重复性和应答敏感性来说 ,CTL L - 2 /IL - 4R较之 T淋巴细胞更为优越 ,该细胞系可用于建立一种较灵敏而稳定的 IL -4测活方法。 ObjectiveTo establish a stable and sensitive assay for the determination of the biological activity of rhIL 4 Methods The characteristic of proliferation response of human peripheral blood T lymphocyte and CTLL 2/IL 4R cell line on rhIL 4 was compared.At same time,some conditions involved in the MTT assay was also compared,such as cell number,culture time,MTT concentration and incubation time, etc .Results The two assays all presented regular response.The lowest detectable biological activity of rhIL 4 determined by human peripheral blood T lymphocyte and CTLL 2/IL 4R cell line was 8 U/ml and 0 73 U/ml respectively.The optimal procedure for MTT assay was also established. Conclusion CTLL 2/IL 4R cell line was found to be superior to T lymphocyte on stability and sensitivity. [
