摘要
以‘热研1号’槟榔幼胚为外植体,采用正交设计法研究不同基本培养基、活性炭及蔗糖等浓度在槟榔胚离体培养中对幼苗生长发育的影响。通过方差分析和差异显著性检验,评价最佳配方组合时,评价指标考虑的优先顺序依次为芽高、最长根长、芽基部直径、根数;MS培养基优于其他几种培养基,能同时满足芽和根的生长需求;活性炭对生根及根的伸长作用显著,4g/L的活性炭浓度时抽生的根最多,长势最好,蔗糖能同时促进根和芽的生长,随添加量的增加,生长指标呈低-高-低的单峰曲线趋势,40g/L蔗糖浓度对芽的生长效果最好。研究得出的最佳配方组合是:MS培养基+4g/L活性炭+40g/L蔗糖。
Using the areca catechu‘Reyan No.1’immature embryo as explants for embryo culture,using orthogonal design method to study the different medium,sucrose and active carbon concentration on Areca catechu embryo in vitro culture on the seedling growth effect.Through the analysis of variance and difference significance test,the best formula of combination evaluation,priority sequence of evaluation index considering was:the bud height→the longest root length→bud base diameter→root number.MS medium was the best in all medium,It can both satisfy the bud and root growth needs.Activated carbon significantly beneficial to take root and growth of root elongation,when used 4g/L of activated carbon,the quantity of root and growth of root grew well,sucrose promoted root and bud growth,with the increase of adding amount of activated carbon,growth index showed low-high-low unimodal curve trend,40g/L sucrose concentration on bud growth best effect.This study concluded that the best formula of combination was:MS medium +4g/L activated carbon +40g/L sucrose.
