摘要
目的 探讨醛固酮合成酶基因CYP11B2在正常与肝纤维化大鼠肝组织中的表达。方法 48只雄性Wistar大鼠,随机分为模型组与对照组。模型组:40% CCl4 油2.5 ml/kg(b.w.皮下注射,每周3次;对照组:橄榄油2.5 ml/kg(b.w.皮下注射。于第4、6、8、10周处死大鼠,光镜下观察肝组织形态学变化。RT-PCR和原位杂交法检测CYP11B2 的表达。结果与结论 原位杂交法及RT-PCR检测显示CYP11B2在纤维化形成时表达增强,提示CYP11B2在纤维化肝脏的储脂细胞中表达增强。
Objective In consideration of the facts that aldosterone synthesis may occur in vascular tissue, brain and liver, and locally produced aldosterone is likely to take an active part in fibrogenesis of the liver, we undertook the present study to identify aldosterone synthase gene-CYP11B2 mRNA expression in normal and fibrotic liver of rats. Methods Forty-eight Wistar rats were randomly divided into 2 groups. In the model group, the rats were injected with 40% CCl4 2.5 ml/ kg(b.w. subcutaneously 3 times a week. The rats in the control group were injected with olive oil only. After 4, 6, 8 and 10 weeks, the animals were sacrificed for morphological examination of the hepatic tissue. By means of RT-PCR and in situ hybridization, the expression of aldosterone synthase gene CYP11B2 mRNA in fibrotic and normal liver were detected. Results and Conclusion In situ hybridization and RT-PCR showed that the expression of CYP11B2 mRNA was up-regulated when fibrogenesis occurred. The expression of CYP11B2 mRNA is up-regulated in fibrotic liver.
