摘要
目的 探测白细胞介素 - 6 (IL- 6 )是否与垂体前叶中的神经纤维的营养效应有关。 方法 应用免疫组织化学和原位杂交技术对去肾上腺大鼠垂体中的 IL- 6表达进行形态观察 ,并试用两种微量 PCR方法对同一组织中的 IL- 6进行蛋白水平和 m RNA水平的定量检测。 结果 免疫组织化学分析 ,阳性物质主要存在于垂体前叶滤泡星形 (FS)细胞中 ,但其含量变化却不甚清晰。PCR定量分析 ,IL- 6在上述两个水平上的表达均有增加 ,且基因激活时间早于术后 2 4h。 结论 IL- 6变化与 ACTH无关 ,可能参与下丘脑 -垂体 -肾上腺 (HPA)轴以外的垂体前叶功能调节的其他路径 。
Objective\ To explore the relationship between IL\|6 and an effect of neurotrophy on the nerve fibres in anterior pituitary. Methods\ To investigate the changes of IL\|6 in the anterior pituitary of adrenalectomized rats,two PCR procedures,Immuno\|PCR(I\|PCR) and RT\|PCR were modified to quantify antigen and mRNA in tiny samples respectively to supplement the morphometry,since morphoanalysis,immunocytochemistry and in situ hybridization seemed not efficient to show quantification. Results\ The most IL6\|IL stainings were found to be gathered mainly in FS cell,and the levels of both protein and mRNA of IL\|6 determined by PCRs rised obviously.Meanwhile,the activation of IL\|6 gene occurred within 24h after adrenalectomy. Conclusion\ This study suggests that IL\|6 might be involved in the regulation of the secretion of anterior pituitary in other way independent of PHA axis,such as neuro\|regulation.\;
