摘要
在还原物质NADPH及硝酸盐还原酶的催化下 ,显微分光光度法测定细胞及其培养液的氮氧化物含量 ,得到满意结果 :Sander灵敏度为 3 8× 10 -6μmoL/cm2 ,变动系数 (CV % ) 3 96 (n =8) ,标准添加回收率 (10 0 1± 4 96 ) % ,浓度 (C)与吸光值 (A)的线性范围为 0 15~ 1 0 0 μmoL/L ,其回归方程为C =- 0 0 18+ 1 0 99A(γ =0 995 8,P <0 0 1) ,在合适的还原条件下 ,酶促的还原效率为 95 9%。
A method was developed for the determination of nitric oxides in cells and its culture medium by means of microspectrophotometry with NADPH and catalyzing of nitrate reductase(NR).As the results,it was found that:the Sander sensibility is 38×10 -6 μmoL/cm2,the coefficient of variation is 396%(n=8),the recovery of using the standard addition method is(100±496)%.A linear calibration curve was obtained in the absorbance~concentration range of (015~100) moL/L,and the regression equation C=-0018+199A(γ=09958,P<001).The reduction efficiency of NO3 in enzyme catalysis was 959%.The present method is good enough for quantitative analysis of nitric oxides in biological samples.
