摘要
目的 观察亚硒酸钠 (sodium selenite,Na2 Se O3)对微波致体外培养的视网膜神经节细胞损伤的防护作用。 方法 体外培养视网膜神经节细胞 ,按加入培养液 Na2 Se O3浓度分为 4组 ,其中 1组为对照组 ,30 m W/ cm2 微波辐射 1h,测定细胞内的脂质过氧化指标及硒浓度 ,末端脱氧核甘酸转移酶介导的生物素化脱氧尿甘三磷酸末端标记法 (Td T- mediated d UTP nick end labeling,TUNEL )检测细胞凋亡。 结果 1× 10 - 7mol/ L Na2 Se O3即可使细胞内超氧化物歧化酶 (superoxide dismutase,SOD)、谷胱甘肽过氧化物酶 (glutatione peroxidase,GSH- Px)活性较未加硒组明显升高 (P<0 .0 5 ) ,丙二醛 (malondialdehyde,MDA)含量显著下降 (P<0 .0 5 ) ,凋亡细胞数量减少 ,1× 10 - 6 mol/ L组和 1× 10 - 5 mol/ L组在抗氧化能力方面优于1× 10 - 7mol/ L 组 ,但这两组间无显著差异。 结论 Na2 Se O3对微波致视网膜神经节细胞的损伤具有一定的保护作用。
Objective [WT5”BZ]To observe the protective effects of Na 2SeO 3 on the damage of retinal neuron induced by microwave. [WT5”HZ]Methods [WT5”BZ]Cultured fluids of retinal neuron were divided into 4 groups,including 1 group of control, according to the concentration of Na 2SeO 3 in cultured fluid and then exposed to 30 mW/cm 2 microwave for 1 hour.The targets of lipid peroxidation and the concentration of selenium in cells were measured.Apoptosis detection was taken by TUNEL detection kit. [WT5”HZ]Results [WT5”BZ]The activity of SOD and GSH Px rised,meanwhile the content of MDA and the amount of apoptosis cells decreased in 1×10 7 mol/L group compared with the group without Na 2SeO 3.The other groups was superior in antioxdant capacity to 1×10 7 mol/L group. [WT5”HZ]Conclusion [WT5”BZ]Na 2SeO 3 might be possessed of the effect of protecting the damage of retinal neuron induced by microwave. [WT5”HZ]
