肿瘤致SD大鼠全骨病变的有效观察指标筛选

摘要 目的 建立可靠的大鼠肿瘤模型 ,针对恶性肿瘤所致的高血钙及肿瘤转移所致的骨病变 ,找出有意义的观察指标。方法 将Walker 2 5 6癌肉瘤细胞接种于大鼠右后肢胫骨上段骨旁肌肉内 ,14天后局部肿瘤生长。随后对血钙、磷、碱性磷酸酶(AKP)、尿羟脯氨酸、全身平均骨密度 5项指标进行测定和分析。结果 与对照组相比 ,肿瘤生长组大鼠血钙明显升高 (P <0 .0 1) ,全身平均骨密度明显降低 (P <0 .0 5 ) ,尿羟脯氨酸排泄增加 ,而血磷、AKP无显著性变化。结论 血钙、全身平均骨密度、尿羟脯氨酸 3项观察指标对判断肿瘤致SD大鼠全骨病变较有意义。 Objective To find out the sensitive index in order to estimate the effects of new anti hypercalcaemia drugs on cancer related hypercalcaemia and bone damage.Methods An in vivo model of carcinosarcoma Walker 256 cell induced cancer was developed and several indexes of bone damage were screened.Walker 256 cell lines were given by intramuscular injection in right leg of rats.Fourteen days after injection,blood was collected to examine serum calcium,phosphate and alkaline phosphatase(AKP).Urine was also collected for 24 hours by metabolic cage in order to examine urinary hydroxyproline.Bone mineral density(BMD)of rat was detected.Results The serum calcium of treated rats increased and BMD decreased,with statistically significant difference compared with control animals( P <0.05).Conclusion Concentration of serum calcium,BMD and urinary hydroxyproline are more sensitive indexes to screen cancer induced bone damages,and are useful parameters to estimate the effects of new anti hypercalcaemia drugs in animal model.
