摘要
目的:通过采用免疫亲和柱-高效液相色谱法来测定花生中的黄曲霉毒素。方法:样品用甲醇+水(70∶30)提取液提取后过滤,滤液经黄曲霉毒素免疫亲和柱净化,洗脱液经N2吹干后,加三氟乙酸和正己烷衍生后,用带有荧光检测器的超高效液相色谱仪测定。结果:四种黄曲霉毒素G2、G1、B2、B1得到很好的分离。检测限分别为:0.2μg/kg、0.4μg/kg、0.2μg/kg、0.4μg/kg。平均回收率在:81.5%~106.2%,相对标准偏差在1.1%~3.5%。结论:该法能使花生中黄曲霉毒素G2、G1、B2、B1得到很好的分离,结果准确。
Objective:To determine aflatoxins in peanuts by using high performance liquid chromatography-fluorescence detector and immuno-affinity column.Methods: The sample was extracted with V(H2O)∶ V(CH3OH)=30∶ 70,then separated by a immuno-affinity column after filteration.The purified solution was concentrated and evaporated to dryness.Aflatoxins were derivatized with TFA and detected and quantitated by HPLC with fluorescence detector.Results: Four aflatoxins G2,G1,B2,B1 were well separated.Detection limits were: 0.2 μg/kg,0.4 μg/kg,0.2 μg/kg,0.4 μg/kg.The average recovery rate: 81.5%~106.2%.The relative standarddeviation:1.1%~3.5%.Conclusion: This method can in make aflatoxins G2,G1,B2,B1 well separated in peanuts in a short time with accurate test result.
