五子衍宗软胶囊体外对精子内Ca^2+浓度的作用 被引量:2

Effects of Wuziyanzong Softgel on the levels of intraspermic Ca^2+

摘要 目的研究五子衍宗软胶囊,仙茅、淫羊藿分别对人精子胞浆Ca^2+浓度的影响。方法115例采自本院男科门诊精液标本,随机分成五子衍宗软胶囊、仙茅、淫羊藿、空白,黄体酮注射液五组,其中黄体酮注射液为阳性对照组。各组分别加入钙离子荧光探针Flu03.AM,用波长488nm激光激发产生荧光,其荧光强度值与Ca^2+浓度正相关,应用流式细胞仪(FCM)测定精子内Ca^2+荧光强度。结果五子衍宗软胶囊组精子内Ca^2+的荧光强度最强,与黄体酮组精子内Ca^2+的荧光强度比较,差异有统计学意义(P〈0.05):黄体酮、五子衍宗软胶囊组分别与仙茅、淫羊藿、空白组比较精子内Ca^2+的荧光强度比较,差异有统计学意义(P〈0.05);仙茅、淫羊藿组与空白组比较,无统计学意义(P〉0.05)。结论五子衍宗软胶囊能够促进精子胞浆Ca^2+浓度的增加,其作用优于黄体酮、仙茅、淫羊藿组。仙茅、淫羊藿不能促进精子内Ca^2+浓度增加。 Objective To investigate the levels ofintraspermic Ca2+ respectively under different concentration of Wuziyanzong Softgel, rhizoma curculiginis and epimedium. Methods Total of 115 specimens were randomly divided into 5 groups such as the Wuziyanzong Softgel group. Rhizoma curculiginis group, Epimedium group, the control group and Progesterone group. The Progesterone group was set as the positive control group. All samples in each group were treated with Fluo3-AM.While Fluo3-AM was passively diffused into the cells, Fluo3 was released and combined with calcium ion. The fluorescence intensity, generated by 488nm wavelength laser, was increased proportionally with increasing concentration of intraspermic Ca 2+. The concentration of intraspermic Ca2+ was mea- sured by flow cytometry(FCM). Results The level ofintraspermic Ca2+ in Wuziyanzong Softgel group was the highest among all the groups, and it had a significant difference compared with that of Progesterone group (P〈0.05). There were significant differences in the levels of intraspermic Ca〉 between Wuziyanzong Softgel group and Rhizoma curculiginis group, Epimedium group or the control group, as well as between Progesterone group and Rhizoma curculiginis group, Epimedium group or the control group (P〈0.05). No significant differences in the levels of intraspermic Ca2+ were found among Rhizoma curculiginis group, Epimedium group and the control group (P〉0.05). Conclusion Wuziyanzong Softgel might promote Ca2+ influx and increase intraspermic concentration of Ca〉, and its effect was better than that of progesterone, rhizoma curculiginis and epimedium. Rhizoma curculiginis and Epimedium had no effects on the concentration of intraspermic Ca^2+.
