摘要
目的探讨不同的方法从母体外周血中分离胎儿有核红细胞的最佳方案。方法分别用密度为10771、11191、10771/11191的细胞分离液密度梯度离心和CD235a、CD71、CD71/CD235a抗体免疫磁珠分离的不同组合,分离后用HbF-FITC荧光抗体标记,流式细胞仪检测,分析各种分离方案从母体外周血中分离胎儿有核红细胞的效果。结果密度为10771细胞分离液与CD71、CD235a、CD71/CD235a免疫磁珠组合,流式细胞仪检测分离的结果为0.27、0.48、1.15,密度为11191细胞分离液与CD71、CD235a、CD71/CD235a免疫磁珠组合,流式细胞仪检测的分离结果为0.18、1.64、5.73,密度为10771/11191的细胞分离液与CD71、CD235a、CD71/CD235a免疫磁珠组合,流式细胞仪检测的分离结果为0.94、2.06、6.21。不同组合之间比较P<0.05,有明显差异。结论用密度为10771/11191的细胞分离液和抗体为CD71/CD235a的免疫磁珠组合分离到的NRBC比例最高。
Objectives:To investigate the best solutions of different methods separating peripheral blood of mother fetal nucleated RBC.Methods: With density respectively for 10771,11191,and 10771/11191 of the cell separation liquid density gradient centrifugation and CD235a,CD71,CD235a/CD71 surface antibody immunomagnetic beads separation of different combination,Use fluorescent antibody HbF-FITC mark,flow cytometric testing after the separation,Analysis the effect of various separation from the peripheral blood of mother separation fetal nucleated RBC.Results:Density for 10771 cell separation liquid and CD71,CD235a,CD71/CD235a immunomagnetic beads combination,the test result of the separation by flow cytometric were 0.27,0.48,1.15 respectively.Density for 11191 cell separation liquid and CD71,CD235a,CD71/CD235a immunomagnetic beads combination,the test result of the separation by flow cytometric were 0.18,1.64,5.73,respectively.Density for 10771/11191 cell separation liquid and CD71,CD235a,CD71/CD235a immunomagnetic beads combination,the test result of the separation by flow cytometric were 0.94,2.06,6.21 respectively.Different combination have obvious difference,P0.05.Conclusion: Tne NRBC proportion for Using density for 10771/11191 of the cell separation fluid and antibody CD71/CD235a immunomagnetic beads combination separation was the highest.
