APPLICATION OF MONOLITHIC POLYCAPILLARIES X-RAY LENS IN MACROMOLECULAR CRYSTALLOGRAPHY

摘要 X光透镜能以较大的收集角收集X光 ,并以准平行光出射 .X光透镜应用于生物蛋白大分子晶体衍射 ,替代单色仪双聚焦镜 ,照射晶体样品的光强提高数倍乃至 1到 2个数量级 ,衍射强度的提高远大于 5倍 ,大大缩短了测试时间 ,分辨能力改善 0 .0 2~ 0 .0 5nm 。 Monolithic polycapillary X ray lens, developed in recent years, has many applications in different fields. Described is a successful application to the X ray diffraction of macromolecular crystallography. By using monolithic X ray lens, the direct beam intensity irradiating on crystal is 1 to 2 order of magnitude higher than the traditional method with double focusing mirrors. The diffracted X ray intensity is increased over 5 times and resolution is improved by 0.02~0.05 nm. Signal to noise ratio is also improved and the time of measurement is reduced. The crystal samples which have to be analyzed by synchrotron radiation before can be investigated by routine laboratory X ray source now.
