泛素-蛋白水解酶复合体通路在小鼠精子变态中的作用 被引量:3

摘要 应用重力沉降法分离睾丸各层细胞 ,通过蛋白聚丙烯酰胺凝胶电泳和蛋白免疫印迹观察各种生精细胞中泛素结合蛋白的表达规律 ,并通过免疫组织化学方法对其进行组织和细胞水平的定位 .结果表明 ,在圆形精子和长形精子细胞中泛素结合蛋白增加 ,在成熟精子中量较低 ,在精原和精母细胞中量微弱 .总蛋白电泳的结果表明 ,在精子变态过程中蛋白的变化以长形精子细胞转变为成熟精子阶段最为剧烈 ,其间许多蛋白消失同时伴随一些新蛋白的出现 .蛋白免疫印迹显示泛素结合蛋白在精子变态过程中大量出现 ;结合总蛋白电泳的结果发现 ,此阶段消失的蛋白多为泛素结合蛋白 .由此可见 ,泛素对一些蛋白的修饰及其降解与精子变态密切相关 . Mouse testis cells are separated by gravitative deposition method. SDS PAGE and Western blot are used to detect the ubiquitin conjugated proteins in different spermatogenic cells. The results indicate there are lot of proteins that degraded from long spermatid to mature sperm. Immunohistochemical analysis shows that ubiquitin conjugated proteins are distributed mainly in round and long spermatid during spermiogenesis in mouse testis crossection. It is suggested that ubiquitin proteasome protein degradation pathway may take great role in mouse spermiogenesis.
