Hoxa9和Shh在神经干细胞定向分化过程中的作用研究

摘要 目的:了解神经干细胞诱导分化所得胆碱能神经元和运动神经元中Hoxa9基因与音猬因子(Shh)的差异表达情况,探讨它们在神经干细胞定向分化过程中的作用。方法:采用实时定量PCR技术,观察Hoxa9基因与音猬因子在胆碱能神经元和运动神经元中的表达情况。结果:Hoxa9在胆碱能神经元中的表达是在运动神经元中的0.169倍(P<0.05)。Shh只在运动神经元中有表达。结论:Hoxa9与Shh在运动神经元和胆碱能神经元之间的表达差异是由于分化时段的不同所导致的。二者在神经干细胞向运动神经元的分化过程中都起着重要的作用。 Objective: To study the differential expression and effect of Hox gene and Shh between the cholinergic neurons and motor neurons which differentiated from neural stem cells. Methods : Use the ABI' s TaqMan MicroR NA Assays realtime PCR to observe the expression of Hox gene and Shh in cholinergic neurons and motor neu rons. Results : The level of Hoxa9 in cholinergic neurons was 0. 169 fold of that in motor neurons ( P 〈 0.05 ), and Shh expressed in motor neurons, but wasn' t detected in cholinergic neurons. Conclusion : The differential ex pression of Hoxa9 and Shh was resulted by differentiation of the different time in motor neurons and cholinergic neurons. Both Hoxa9 and Shh play an important role in differentiation from neural stem cells to motor neurons.
