摘要
目的:探讨8周游泳运动对注射D-半乳糖致雄性衰老大鼠空间学习记忆能力、血清睾酮及伏隔核c-fos表达的影响。方法:30只雄性SD大鼠随机分为正常对照组(C)、衰老模型对照组(AC)和衰老模型训练组(AT),每组10只。检测血清睾酮,利用八臂迷宫测试大鼠空间学习记忆能力,采用免疫组织化学结合图像半定量方法对伏隔核c-fos神经元的数量、面积及灰度进行测量和分析。结果:1)与正常对照组比较,衰老模型对照组大鼠血清睾酮水平显著降低(P<0.01),VME、RME及TE均显著增加(P<0.01);与衰老模型对照组比较,衰老模型训练组大鼠血清睾酮水平增高(P<0.05),VME、RME及TE均显著减少(P<0.05,P<0.01);2)与正常对照组比较,衰老模型对照组Nac c-fos免疫阳性细胞数量和阳性产物面积均减少(P<0.01),灰度没有显著差异(P>0.05);与衰老模型对照组比较,衰老模型训练组Nac c-fos免疫阳性细胞数量和阳性产物面积均增加(P<0.05),灰度增加,但没有显著差异(P>0.05)。结论:游泳运动可提高老年大鼠的记忆能力,机理可能与长期游泳运动提高老年大鼠的血清睾酮水平及增强大脑伏隔核c-fos的表达有关。
Objective : To explore the effects of the 8 - week swimming exercise on learning and memory, the level of serum testosterone, and expression of c-fos within nucleus accumbens (Nac) of brain in the aging male rats by D-galactose. Methods: Thirty SD aged rats were chosen and randomly divided into 3 groups: the control group (C), the aging model control group (AC), the aging model training group (AT). Radial maze was used to as- sessed each animal' s capability of learning and memory, the level of serum testosterone was examined, and c-fos expression within Nac was detected and analyzed by using ABC immunohistochemical technique and semi quantita- tive. Resuhs:l ) Compared with the C group, the level of testosterone of blood plasma decreased (P 〈 0.01 ) , VME, RME and TE significantly increased (P 〈 0.01 )in the AC group; Compared with the AC group, the level of testosterone of blood plasma increased ( P 〈 0. 05 ) , VME, RME and TE significantly decreased ( P 〈 0.05 ) in the AT group; 2) Compared with the C group, the amount and area of c-fos within Nac obviously decreased ( P 〈 0.05 ) , the gray degree was not significant different ( P 〉 0.05 ) in the AC group ; Compared with the AC group, the amount and area of c-fos within Nae obviously increased ( P 〈 0.05 ) , the gray degree increased but it was not significant different ( P 〉 0. 05 ) in the AT group. Conclusion : The swimming exercise may enhance the learning and memory ability of the aging rats, its mechanism might be that swimming exercise increase the level of serum testosterone and reinforced expression of c-fos within Nac at the same time.
