摘要
目的观察野牡丹提取物对血栓形成、血小板聚集及血小板cAMP、cGMP和一氧化氮(NO)含量的影响。方法在颈外静脉注射给药5min后,用血栓生成仪观察野牡丹提取物对大鼠颈总动脉血栓形成的影响,测定5min平均堵塞率;全血与药物孵育5min,用全血血小板聚集仪观察全血血小板聚集情况;用ELISA法检测血小板cAMP和cGMP含量;用硝酸还原酶法观察血小板释放NO情况。结果野牡丹提取物5mg/kg促进大鼠颈总动脉血栓形成,堵塞率为(86.56±1.54)%。200、400、800mg/L野牡丹提取物均能诱导全血血小板聚集,并减少血小板cAMP、cGMP和NO生成。结论野牡丹提取物可促进大鼠血栓形成和血小板聚集,并能减少血小板cAMP、cGMP和NO含量。
Objective To study the effect of Melastoma candidum extraction (MCE) on thrombosis, platelet aggregation, and platelet cAMP, cGMP and nitric oxide (NO) concentration. Methods The thrombosis of the common carotid artery and average 5-minute blockage rate were determined by thrombotic detector after 5 min of MCE injection via external jugular vein in rats. After whole blood was incubated with MCE for 5 min, the platelet aggregation and platelet levels of cAMP, cGMP and NO were detected by aggregometer, ELISA and nitrate reductase assay, respectively. Results 5 mg/kg MCE increased the thrombosis of the common carotid artery, and the blockage rate was (86.56±1.54)%. 200, 400 and 800 mg/L MCE induced the platelet aggregation, and decreased the platelet contents of cAMP, cGMP and NO. Conclusion MCE can reinforce the thrombosis and platelet aggregation, but reduce the platelet production of cAMP, cGMP and NO.
