摘要
目的 :构建一个含有 HBV X(HBx)基因和它的调控序列的重组质粒。方法 :通过 PCR法获取目的基因 ,将目的基因与 p GEM- 5 zf(+) T载体相连组成重组质粒 ,转入大肠杆菌 JM10 9株后获得重组克隆。并分别通过PCR、限制性内切酶酶切图谱和序列分析 3种方法对重组质粒进行鉴定。结果 :PCR、限制性内切酶酶切图谱和序列分析均证实所获重组质粒中含有目的基因。结论 :得到了含有 HBx结构基因及其上游调控序列的重组克隆 ,为进一步研究
Objective: Our purpose was to construct a reconstructed plasmid carrying hepatitis B virus(HBV) X structure gene and its regulation sequence. Methods: The target gene was acquired by PCR method and pGEM 5zf(+) T vector was linked to the target gene. The reconstructed plasmid (pGBX) was identified by PCR, restricted enzyme map, and sequence analysis. Results: The reconstructed plasmid was proved to carry the target gene by PCR, restricted enzyme map, and sequence analysis. Conclusion: We acquired a reconstructed plasmid that carries HBx structure gene and its regulation sequence. This experiment will build up a foundation for the following study of HBx's function.
