蛭龙胶囊联合坎地沙坦对早期DN大鼠肾脏保护作用的实验研究

Protective effect of Zhilong capsule combining candesartan on early renal fibrosis in DN rats

目的：通过观察4组DN模型大鼠（B组模型组、C组给予蛭龙胶囊治疗、D组给予坎地沙坦治疗、E组给予蛭龙胶囊联合坎地沙坦治疗）及正常对照组（A组）之间血浆TXB2、6-keto-PGF1α及肾小球形态学变化情况，探讨蛭龙胶囊对早期DN大鼠肾脏保护作用。方法：60只2～3月龄、尿蛋白阴性的SPF级雄性SD大鼠随机分为4组，每组15只，另外10只为正常对照纳入研究，建立DN模型大鼠，8周后分别测定各组大鼠血浆血栓素B2（TXB2）和6-酮-前列腺素F1α（6-Keto-PGF1α）含量及评估显微镜下肾脏肥大指数，并进行多组间单因素方差分析及SNK-q检验。结果：研究发现C组大鼠TXB2降低、6-keto-PGF1α升高，与除E组外其他各组比较差异均有统计学意义（P〈0.05）；4组模型大鼠肾脏肥大指数均高于A组（P〈0．01）；C-E组大鼠肾脏肥大指数较B组明显下降（P〈0.05），E组下降更明显（P〈0．01）；与C组比较，D组大鼠肾脏肥大指数差异无统计学意义（P〉0．05），但E组差异有统计学意义（P〈O．05）。结论：蛭龙胶囊有抑制DN模型大鼠肾间质纤维化及肾脏肥大的作用，且联合坎地沙坦治疗后该效应更明显。

Objective： To discuss the effect of Zhilong Capsule on TXB2、6-keto-PGF1α, and the morphologic variations in rats of early diabetic nephropathy, with four groups diabetic nephropathy rats （group B as model, group C as therapy with Zhilong capsule, group D as therapy with Candesartan, group E as therapy with Zhilong eapule combining with Candesartan, group A as normal ）. Methods： 65 SPF grade, 2-3 months old, proteinuria negative, male Wistar rats, were made the early stage DN model,and another 10 rats were taken as normal group in which the level of TXB2 and 6-keto-PGF1α were detected in every group after 8 months therapy, and renal hypertrophy index（KI） was evaluated under the microscope, and analysed the data by one factor analysis of variance. Results： Compare with other groups except for group E,the level of TXB,. decreased and the contents of 6-keto-PGF1α increased in group C in which there was a significant difference （P〈0.01）, and the KI of four groups were obviously higher than that in group A （P〈0.01）.Compared with group B, from group C to E were obviouslydeclined （P〈0.05）, the group E was more obvious（P〈0.01 ）. Compared with group C, the KI of group D had no significant difference（P〉0.05）, group E had significant difference （P〈0.05）. Conclusion： Zhilong capsule can protect the renal interstitial fibrosis and renal hypertrophy of DN rats in which Zhilong capsule combining with Candesartan can make beter effection.
