一种水解血小板膜糖蛋白GPIb的眼镜蛇毒组分的分离纯化与鉴定 被引量:1

摘要 目的 建立从中华眼镜蛇毒中分离、纯化可水解血小板膜糖蛋白Ib(plateletmembraneglycoproteinIb,GPIb)组分的方法。方法 肝素亲和层析,分子筛层析,SDS聚丙烯酰胺凝胶电泳(SDSPAGE)。结果 经HeparinSehparoseCL6B亲和层析及SephadexG150分子筛层析,从中华眼镜蛇毒纯化出可降解纤维蛋白原和GPIb的活性组分,回收率为0-88%,SDSPAGE证实此组分为相对分子量约47100的单肽链蛋白。结论 此方法成功地从中华眼镜蛇毒中纯化出了水解GPIb的组分,具有高效、简便的特点。 AIM To establish a purification method for the fraction in Chinese cobra venom ( Naja naja atra ) which degrades platelet glycoprotein Ib (GPIb) METHODS Heparin affinity chromato graphy, gel filtration, SDS polyacrylamide gel electrophoresis (SDS PAGE) RESULTS By means of Heparin Sepharose CL 6B affinity chromatography and gel filtration on Sephadex G 150 column, a fraction from the venom of Naja naja atra was purified to homogeneity by the citerion of SDS PAGE with a recovery of 0 88% This fraction can degrade fibrinogen and GPIb SDS PAGE showed it was a single polypeptide with a molecular mass of 47 100 CONCLUSION The method is simple and effective for the purification of the fraction in Chinese cobra venom which bears proteolytic activity against GPIb
