5-氯水杨酰肼三苯基氯化锡体外抗肿瘤作用研究 被引量:1

Study on the Anti-Tumor Effects of(Ph_2Sn)_2(HL_9)_2(C_7H_5O_2N_2)Cl in vitro

摘要 研究5-氯水杨酰肼三苯基氯化锡(LPY-3)体外抗肿瘤作用及作用机制。采用MTT法检测LPY-3对HO-8910、Hela、A549等6种肿瘤细胞的生长抑制作用;用Western Blotting法检测LPY-3对HO-8910细胞中凋亡相关信号分子及DNA断裂标志蛋白γ-H2AX的诱导作用;用DNA凝胶电泳检测LPY-3对pBR322质粒的DNA断裂作用。结果表明,LPY-3对HO-8910、Hela、A549等细胞有显著的增殖抑制作用,IC50值在0.12~0.26μmol/L浓度范围内;能够诱导HO-8910细胞中凋亡相关信号分子Caspase-9、Caspase-3、Cyt C、PARP剪切,Bcl-2表达降低,γ-H2AX表达升高;对pBR322质粒有DNA断裂作用。 The anti-tumor effects and mechanisms of (Ph2Sn)2(HL9)2(CTHSO2N2)Cl in vitro were in vestigated. MTT assay was used to detect the cytotoxic effect of LPY-3 on six kinds of tumor cell lines. Western Blotting was used to identify the inducements HO-8910 and the symbol protein γ-H2AX about DNA of LPY-3 to the apoptosis signaling molecules in cleavage. Agrose gel electrophoresis was used to detect the cleaving effect of LPY-3 on pBR322. The results indicated that LPY-3 significntly inhibited the growth of these six kinds of tumor cell lines, and their IC50 fall in the 0. 12-0. 26 μmol/L range. LPY-3 can trigger the activation of apoptosis signaling molecules (caspase-3, caspase-9,Cyt C and PARP ), decrease the expression of Bcl-2 and oppositely advance the expression of γ-H2AX. LPY-3 can cause the DNA cleavage of pBR322.
