电针对多囊卵巢综合征大鼠干细胞因子及肿瘤坏死因子-α表达的调节 被引量:18

摘要 目的观察电针干预对多囊卵巢综合征(PCOS)大鼠卵子成熟度的影响及对干细胞因子(SCF)和肿瘤坏死因子-α(TNF-α)表达的调节,并探讨其机理。方法 24日龄SD雌性大鼠30只,10只为空白组,予普通饲料喂养,不予任何干预;20只予高脂饲料喂养,予脱氢表雄酮造模,造模成功后大鼠分为电针组与模型组。电针组予电针干预,取穴"肾俞"、"气海"、"内关"、"足三里"、"三阴交"、"子宫",采用疏密波,每日1次,每次30 min。模型组不予任何处理。观察3组大鼠成熟卵子数量、血清激素水平、颗粒细胞悬液SCF及TNF-α的表达。结果对PCOS大鼠模型进行成功造模。模型组与电针组卵巢湿重明显高于空白组(P<0.05)。空白组雌二醇明显高于模型组(P<0.05),略高于电针组,但差异无统计学意义(P>0.05);模型组睾酮明显高于电针组、空白组(P<0.05,P<0.01),后两者比较无明显差异。电针组成熟卵子数及优卵率高于模型组,但均低于空白组,且模型组与空白组比较差异有统计学意义(P<0.05)。3组SCF及TNF-α比较差异有统计学意义(P<0.01)。电针组PCOS大鼠恢复动情周期。结论电针刺激可通过改善卵巢局部微环境SCF与TNF-α水平来刺激PCOS大鼠卵子的发育及成熟。 Objective: To investigate the effects of electro-acupuncture (EA) treatment on eggs maturity and the level of SCF and TNF-a in rat of polycystic ovarian syndrome (PCOS), and to explore the possible mechanism associated. Methods In thirty 24-day-old female SD rats, 10 for blank group were given ordinary feed and without any intervention, 20 were given high fat feed and Dehydroepiandrosterone to made PCOS model. PCOS rats were divided into EA group and model group. EA group received EA treatment at BL23 (Shenshu), RN6 (Qihai), PC6 (Neiguan), ST36 (Zusanli), Sp6 (Sanyinjiao), EX-CA1 (Zigong). The difference of the quantity of mature eggs, the level of SCF and TNF-a of the three groups was observed. Results It was succeed in malting the model of PCOS in rats. Compared with that in the blank group, the weight of ovary in model group and EA group were obviously higher. The level of E2 in blank group was significantly higher than that of model group (P〈0.05), but had no difference with EA group (P 〉0.05). The level of T in model group was significantly higher than those of blank group and EA group (P 〈0.05). The amount of mature eggs and the rate of the high quality egg in EA group were higher than those of model group, but were lower than those of blank group. The level of SCF in blank group was significantly higher than the model group and EA group (P〈0.01). The level of TNF-a in blank group was significantly lower than the two groups (P〈0.01). The rats in EA group resumed the estrous cycle. Conclusion EA can accelerate the development and maturation of the eggs in PCOS rats. It might be related with the role of EA on adjusting the level of SCF and TNF-a in local tance of ovary.
