肺纤方对肺纤维化大鼠肺组织自由基损伤的影响 被引量:6

Effect of Formula for Lung Fibrosis on Free Radical in Pulmonary Tissue of Pulmonary Fibrosis Rats

摘要 目的观察肺纤方对博来霉素所致肺纤维化大鼠肺组织自由基损伤的影响,探讨其抗肺纤维化的作用机制。方法 144只健康Wistar大鼠随机分成空白对照组、模型组、阳性对照组及肺纤方高、中、低剂量组,每组24只。除空白对照组外,按Szapiel法建立模型,次日灌胃给药,肺纤方高、中、低剂量组(3.2、1.6、0.8 g/kg)均为10 mL/kg,阳性对照组予醋酸泼尼松混悬液(6 mg/kg),空白对照组与模型组予等量生理盐水,1次/d,给药周期21 d。分别在造模7、21 d后分2次取材,前后期各随机杀检10只。测定肺组织谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)的含量。结果与模型组比较,肺纤方高、中剂量组均可显著升高肺纤维化大鼠肺组织中GSH-Px和SOD的含量(P<0.05);其中第21日时,肺纤方高剂量组升高SOD的作用最为显著(P<0.01)。与模型组相比,肺纤方高、中剂量组均可显著降低肺纤维化大鼠肺组织中MDA和NO的含量(P<0.05);其中第21日时,肺纤方高剂量组降低NO含量的作用最为显著(P<0.01)。结论肺纤方能不同程度调节大鼠肺纤维化模型体内的自由基代谢,从而缓解肺泡炎程度,减轻大鼠肺纤维化病变,且呈现一定的量效关系。 Objective To observe the effect of Formula for Lung Fibrosis on free radical in pulmonary tissue in rat model of pulmonary fibrosis induced by Bleocamicina. Methods A total of 144 healthy Wistar rats were divided into six groups randomly, which were blank control group, model group, control group, and high-, medium-, low-dose treatment group of Formula for lung fibrosis, 24 rats in each group. The model was made with Szapiel's method, and the next day rats were given drug with intragastric administration. The three treatment groups were given high-, medium- and low-dose of Formula for Lung Fibrosis (3.2, 1.6, 0.8 g/kg, which were as 10 mL/kg), control group was given suspl, prednisone acetate (6 mg/kg, which was as 10 mL/kg), and blank control group and model group were given normal saline once a day for 21 days. Samples were drawn at the 7th day and 21st day, 10 rats respectively. GSH-Px, SOD, MDA and NO in lung tissue were detected. Results Compared with model group, GSH-Px and SOD increased remarkably in high- and medium-dose treatment group of Formula for Lung Fibrosis (P ~0.05). At the 21st day, the increased SOD was the highest in high dose treatment group of Formula for Lung Fibrosis (P^0.01). And compared with model group, MDA and NO decreased remarkably in high- and medium- dose treatment group of Formula for Lung Fibrosis (P ~0.05). At the 21st day, the decreased NO was the lowest in high dose treatment group of Formula for Lung Fibrosis (P^0.01). Conclusion Formula for Lung Fibrosis can regulate the metabolism of free radical in rat model of pulmonary fibrosis at different extent. It can prevent rat model induced by Bleocamicina from injury of lung, relieve airsacculitis, and lighten pathological changes of pulmonary fibrosis with dose-effect relationship.
