小归芍化浊解毒方对胃癌前病变大鼠表皮生长因子受体及血管内皮生长因子表达的影响

Effect of Eliminate Turbid Fluid and Remove Toxicity of Xiaoguishao on EGFR and VEGF in Rats with Gastric Carcinoma Precancerosis

目的探讨小归芍化浊解毒方对实验性胃癌前病变的作用和机制。方法按随机数字表把100只大鼠随机分成为A组(空白组)、B组(模型组)、C组(中药大剂量组)、D组(中药小剂量组)、E组(对照组),每组20只。除空白组外,其余各组采用N-甲基-N-硝基-N-亚硝基胍(MNNG)配合饥饱失常诱导造模。造模后A组正常喂养,B组予蒸馏水灌胃,C组按2 mL/kg予小归芍化浊解毒方大剂量混悬液灌胃,D组按2 mL/kg予小归芍化浊解毒方小剂量混悬液灌胃,E组予胃复春混悬液灌胃。观察大鼠胃黏膜的组织病理变化,测定胃组织表皮生长因子受体(EGFR)及血管内皮生长因子(VEGF)表达。结果与胃复春组比较,小归芍化浊解毒方大剂量组能有效改善胃黏膜肠化和异型增生(P<0.05),小归芍化浊解毒方大、小剂量组能明显降低VEGF的表达水平(P<0.05),小归芍化浊解毒方大剂量组能明显降低EGFR的表达水平(P<0.05)。结论小归芍化浊解毒方可能通过降低VEGF及EGFR的表达,逆转胃癌前病变。

Objective To explore the mechanism of eliminate turbid fluid and remove toxicity of Xiaoguishao on experimental gastric carcinoma precancerosis. Methods According to the random number table, 100 normal rats were divided into group A （blank control）, group B （model）, group C （TCM with large dosage）, group D （TCM with small dosage）, group E （control）, with 20 rats in each group. By using N-methyl-N-nitro-N-nitrosoguanidine and method of irregular diet, all rats but blank control group were induced to gastric carcinoma precancerosis models. After creation of models, group A were feed normally, group B, C, D and E were intragastrically administrated with distilled water, suspension of eliminate turbid fluid and remove toxicity of Xiaoguishao with large dosage 2 mL/kg, suspension of eliminate turbid fluid and remove toxicity of Xiaoguishao with small dosage 2 mL/kg, suspension of Weifuchun respectively Then histopathology of gastric tissues was observed. Meanwhile, the levels of EGFR and VEGF in gastric tissues were tested. Results Compared with group E, the histopathology in gastric tissues of group C were significantly ameliorated （P〈0.05）, VEGF of group C and D were significantly reduced （P〈0.05）, EGFR of group C was significantly reduced （P〈0.05）. Conclusion By means of reducing the levels of VEGF and EGFR, reversal effect of eliminate turbid fluid and remove toxicity of Xiaoguishao on gastric carcinoma precancerosis was produced.
