环孢素A治疗再生障碍性贫血体外药敏试验的初步研究

A study on cyclosporin A sensitivity test in aplastic anemia

目的 建立一种有利于提高环孢素A(CsA)治疗再生障碍性贫血 (再障 )疗效的体外药敏试验方法。方法 用体外集落培养法对 14例再障患者进行CsA体外敏感试验并研究其与临床疗效的关系。结果 体外 /体内 (S/R) (S :敏感 ,R :耐药 ) :S/S 7例 ,R/R 6例 ,R/S 1例。与临床总符合率为 92 9% ,阳性符合率 10 0 % ,阴性符合率 85 7% ,试验敏感性 87 5 % ,特异性 10 0 %。结论 CsA体外敏感试验与临床疗效有很好的相关性 。

Objective To find a method of testing cyclosporin A (CsA) sensitivity for aplastic anemia (AA) in vitro Methods The sensitivity of AA to CsA was estimated in 14 patients with AA by colony formation assay in vitro The relationship of sensitivity test with clinical efficacy was investigated also Results 7 patients were found to be sensitive both in vitro and in vivo, 6 to be resistant both in vitro and in vivo and the remaining one to be resistant in vitro, but sensitive in vivo The general, positive and negative coincident rates of in vitro with in vivo and the specificity and sensitivity were 929%, 100%, 857%, 875% and 100% respectively Conclusion This method is a good single drug test for the therapy of AA
