摘要
目的 探讨肝硬化门静脉高压症患者肝脏组织α1肾上腺素受体亚型mRNA的表达。 方法 应用半定量逆转录聚合酶链反应检测了 12例乙型肝炎后肝硬化门静脉高压症患者和 15例无高血压 ,无肝脏疾患的胆石症或胃肠道肿瘤患者肝脏组织α1肾上腺素受体亚型mRNA的表达。在同一体系中同时逆转录并扩增α1肾上腺素受体亚型特异性片段和内参GAPDH片段 ,以二者的积分光密度比值作为该受体亚型的mRNA相对表达量。 结果 肝脏组织中α1a肾上腺素受体亚型的相对表达量 :对照组 0 86± 0 38显著高于肝硬化组 0 2 6± 0 12 (P <0 0 1) ;α1b肾上腺素受体亚型的相对表达量 :对照组 0 2 3± 0 10显著高于肝硬化组0 0 3± 0 0 1(P <0 0 1)。对照组及肝硬化组肝组织中α1a肾上腺素受体亚型的表达量均显著高于α1b肾上腺素受体亚型 (P<0 0 1)。对照组及肝硬化组肝组织中均无α1d肾上腺受体亚型的表达。 结论 正常及肝硬化肝组织中均以α1a肾上腺素受体亚型的表达为主 ,α1b肾上腺素受体亚型次之 ,无α1d肾上腺受体亚型的表达。肝硬化肝组织中α1a和α1a肾上腺素受体亚型表达减少是造成肝硬化时肝脏α1肾上腺素受体蛋白减少的重要原因 ,对门静脉高压症的形成和维持起重要作用。
Objective[WT5”FZ] To investigate the expression of mRNA of α 1 adrenoceptor in the hepatic tissues of cirrhotic patients with portal hypertension. [WT5HZ]Methods[WT5”FZ] Semi quantitative reverse transcription and polymerase chain reaction (RT PCR) was used in 12 hepatitis virus B related cirrhotic patients with portal hypertension and 15 controls who had no history of portal hypertension. In these patients, routine liver biopsy was performed. Special α 1 adrenoceptor subtype product and internal standard GAPDH product were both amplified by RT PCR. The ratio of their integral optical density (IOD) was calculated as the relative expression of the α 1 adrenoceptor subtypes. [WT5HZ]Results[WT5”FZ] The relative mRNA expression of α 1a adrenoceptor subtype in the hepatic tissues was significantly lower in the cirrhotic patients (0 26±0 12) than in the controls(0 86±0 38, P <0 01). The relative mRNA expression of α 1b adrenoceptor subtype in the hepatic tissues was also significantly lower in the cirrhotic patients (0 03±0 01) than in the controls (0 23±0 10, P <0 01). Neither the cirrhotic patients nor the controls showed α 1d adrenoceptor subtype in the hepatic tissues. The relative mRNA expression of α 1a adrenoceptor subtype was significantly higher than that of α 1b adrenoceptor subtype in the hepatic tissues of both cirrhotic patients and controls ( P <0 01). [WT5HZ]Conclusions[WT5”FZ] The α 1a adrenoceptor mRNA expression predominats in both normal and cirrhotic hepatic tissues. The expression of α 1b adrenoceptor is less marked and there is no expression of α 1d adrenoceptor. The decreased expression of mRNA of α 1a and α 1b adrenoceptor may underlie the decrease of α 1 adrenoceptor proteins often seen in the hepatic tissues of cirrhotic patients, and may play an important role in the pathogenesis and maintenance of portal hypertension.
