石吊兰素降压效应及其机制的实验研究 被引量:4

摘要 目的本实验通过在侧脑室注射石吊兰素观察大鼠血压的变化,并探讨其初步机制。方法将48只SD大鼠随机分为四组,分别为生理盐水对照组(25μL);石吊兰素组(25μg/25μL);酚妥拉明+石吊兰素组(预先注射酚妥拉明25μg/20μL,40min后注射石吊兰素25μg/25μl);美托洛尔+石吊兰素组(预先注射美托洛尔20μg/20μL,40min后注射石吊兰素25μg/25μL);30分钟后观察血压的变化。结果生理盐水对照组,血压无明显改变;石吊兰素组(25μg/25μL),血压下降,收缩压下降幅度为20.2%,舒张压下降幅度为17.1%,与对照组比有显著性差异(P<0.05);酚妥拉明+石吊兰素组,30min后血压明显升高(P<0.05);美托洛尔+石吊兰素组,30min后血压下降(P<0.05)。结论右侧脑室注射石吊兰素具有降低动脉血压的作用,且第40min时下降最明显;石吊兰素降低收缩压的作用较强于降低舒张压,其机制可能为石吊兰素激动中枢α肾上腺素能受体(α-AR)的作用强于中枢β肾上腺素能受体(β-AR)的作用。 Objective To study the effect of injecting Nevadensin into the lateral ventricle on blood pressure in rats and its prelimi nary mechanism. Methods ①The rats were divided into four groups: the saline control group (25μg/25μL), the Nevadensin group (25 μg/25μL), the Pbentolamine and Nevadensin group (injected the 25μg/20μL Phentolamine in advance, and then injected 25μg/25μL Nevadenism 40 minutes later) and the Metoprolol and Nevadensin group (injected the 20μg/20μL Metoprolol in advance, and then injected the 25μg/25μL Nevadenism 40 minutes later). The changes of the blood pressure of the groups were observed 30 minutes later. Results ①The saline control group had no distinct change. The blood pressure of Nevadensin group (25μg/25μL) dropped (the systolic pressure dropped by 20.2% and the diastolic pressure dropped by 17.1%). ②The blood pressure of the Phentolamine and Nevadensin group Raised af ter 30 minutes. The blood pressure of the Metoprolol and Nevadensin group dropped after 30 minutes. Conclusions ①Nevadensin injected into the right lateral ventricle has an effect of lowering the arterial blood pressure and it has the most distinct drop in the 40th minute. ②The effect of lowering systolic pressure resulted by Nevadensin is more effective in lowering diastolic pressure. The machanism of the effect may be that Nevadensin has a greater extend to effect on the α-adrenergic receptor in the centrum than on the β-adrenergic receptor.
