桑树细菌性枯萎病病菌致病性测定的人工接种方法 被引量:7

摘要 为了寻找一种简便、有效测定桑树细菌性枯萎病病菌(Eenterobacter cloacae)致病性的方法,通过离体注射菌液(注射法)、离体牙签蘸菌苔刺伤(牙签法)、离体泡菌液、苗期菌液灌根和苗期泡菌液共5种人工接种方法测定病原菌对桑树的致病性。结果表明,5种人工接种方法仅有注射法和牙签法能成功将病原菌接种至桑树嫩梢,其中牙签法试验组桑树嫩梢的发病率和病情指数最高,分别达到78.99%和54.17,与注射法相比较达到差异极显著水平。因此,简便、有效的牙签法可以作为室内测定桑树细菌性枯萎病病菌致病性及桑树抗细菌性枯萎病育种材料鉴定的首选人工接种病原菌的方法。 In order to find a simple and efficient method for determining the pathogenicity of Enterobacter cloacae that causes mulberry bacterial wilt,five inoculation methods,namely in vitro needle injecting of bacterial liquid(injection method),in vitro toothpick picking and pricking of bacterial colony(toothpick method),in vitro soaking in bacterial liquid,soaking of wounded seedling root in bacterial liquid,and soaking of seedling in bacterial liquid,were used for artificial inoculation of the pathogenic bacteria into mulberry.The results showed that,among the five methods,only injection method and toothpick method could successfully inoculate the pathogenic bacteria into mulberry top shoots and toothpick method had higher disease incidence(78.99%) and disease index(54.17),showing extremely significant difference compared to injection method.Therefore,the simple and efficient toothpick method can be the artificial inoculation method of first choice for indoor pathogenicity determination of Enterobacter spp.and for identifying mulberry breeding materials resistant to bacterial wilt disease.
