摘要
目的观察大鼠蛛网膜下腔出血(subarachnoidhemorrhage,SAH)后N-甲基-D-天门冬氨酸受体亚单位NR2B在脑组织中的表达变化。方法42只成年健康雄性SD大鼠,随机分为正常组、对照组及SAH后1d(D1)、3d(D3)、5d(D5)、7d(D7)、14d(D14)共7组,各6只。SAH模型制作:经大鼠自体尾动脉(股动脉)非肝素化动脉血注入视交叉池,分别在蛛网膜下腔出血后1、3、5、7、14d处死,取大脑皮层、海马、小脑做标本,Western blot及免疫组化测定NR2B。结果对照组与正常组相比差异无统计学意义;SAH后1d组NR2B蛋白水平明显降低(P〈0.01),SAH后3、5、7d组NR2B蛋白水平降低(P〈0.05),SAH后14d组NR2B蛋白水平与正常组相比差异无统计学意义。结论NR2B在SAH后24h明显下降,之后逐渐上升,至14d时基本恢复正常。
Objective To observe the expression changes of NR2B subunit of the N-methyl-D-aspartate receptor in the brain tissue after subarachnoid hemorrhage ( SAH ) . Methods 42 male Sprague Dawley rats were randomly divided into seven group, i.e.normalgroup, control group, 1 days (D1) , 3 days (D3) , 5 days (D5) , 7days (D7) , 14 days ( D 14 ) after SAH. All SAH animals were subjected to injection of 0.3ml fresh arterial non-heparinized blood into prechiasmatic cistern. Brain samples ( cerebral cortex, hippocampus, cerebellum ) were extracted in 1, 3, 5, 7, 14days after SAH. The expressions of NR2B were measured by western blot and immunohistochemistry. Results NR2B proteins were low-regulated significantly following SAH ( P〈0.01 ) .There was no statistically significant difference between normal group and control group ( P〉0.05 ) .The decrease activity of NR2B in 1 day after SAH was markedly reduced in animals of the normal group ( P〈0.01 ) . Compared with the normal group, D3, D5, D7, D14 group had reduced ( P〈0.05 ) , D14 group had no significant difference ( P〉0.05 ) . Conclusion NR2B at 24 hours after SAH decreases significantly, then gradually rises, and returns to normal in 14 days.
