丹参萃取液对肌成纤维细胞表达TGF—β1、α—SMA蛋白的影响 被引量:1

摘要 【摘要】目的进一步探讨丹参萃取液在预防良性胆管狭窄中的作用。方法制作犬胆管损伤修复模型,采用免疫组化SP法对在胆道愈合过程不同时期组织中的转化生长因子-β1(transfotruing growth faetorDl,TGF-β1)、α平滑肌肌动蛋白(α smoothmlls museactin,α-SMA)表达和分布进行研究;通过原代肌成纤维细胞(MFB)的分离及培养,丹参萃取液干预MFB24h后,Westernblot检测TGF-β1、α—SMA在纤维细胞中的表达变化。结果犬胆管损伤修复模型高表达TGF-β1、α-SMA;丹参萃取液作用于MFB后,TGF-β1、α-SMA表达下降。结论丹参萃取液作用于MFB后,TGF-β1、α-SMA等炎症因子分泌减少,胆道瘢痕形成减少,其对预防良性胆管狭窄有一定作用。 Objective To explore the role of salvia extraction fluid in the prevention of benign biliary stricture. Methods A dog model ment of repair of bile duct injury was estahlisbed. The expressions and distribution of TGF-βl and α-SMA in tissues at different phases of the healing process were determined by immunohistoehemical SP stuining. Primary generationment of myofibroblasts was isolated and then cultured. 24 hours 'after the intervention of myofibroblasts by salvia extraction fluid, expressions of TGF-βl and α- SMA were determined by Western blot. Results TGF-βl and α-SMA were highly expressed in the dog model. Salvia extraction fluid acted on the myofibroblasts, resulting in a decrease in the expressions of TGF- βl and α-SMA. Conclusions Salvia extraction fluid acts on myofibroblasts, leading to a decline in secretion of TGF-βl and α-SMA and a decrease in formation of bile duct scars. It plays a role in the the prevention of benign biliary stricture.
