麦门冬汤对放射性肺损伤大鼠肺组织TGF-β、TNF-α表达的影响 被引量:5

摘要 目的探讨麦门冬汤对放射性肺损伤大鼠肺组织TGF-β、TNF-α表达的影响。方法将72只健康成年清洁级雌性Wistar大鼠随机分为空白对照组(A组)24只,单纯照射组(B组)24只,照射加中药组(C组)24只,A、B2组以蒸馏水2ml/200g灌胃1周后,C组以每日2ml/200g(1.6g/200g)灌胃服用中药1周,开始照射,照射期间,每周测体重1次,至照射结束。分别在完成10次照射后(第5周),以及在照射开始后的第12、26周,检测大鼠检测3组大鼠肺组织TGF-β、TNF-α表达。结果 TGF-β蛋白表达:B组大鼠TGF-β的表达于照射第5周时达到高峰;在各时间点,A、C组大鼠TGF-β表达均低于B组(P<0.01)。TNF-α表达:B组大鼠肺组织TNF-α的表达在第5周时达到高峰,C组在第5周时明显抑制了TNF-α表达,在第5周、12周、26周,C组大鼠TNF-α表达均明显低于B组(P<0.01)。结论中药麦门冬汤具有较好的预防放射性肺损伤功效,其作用机制与阻抑肺组织TGF-β、TNF-α表达有关。 Objective To investigate the effect of Maimendong decoction on the expressions of transforming growth factor( TGF-β ), tumor necrosis factor-alpha ( TNF-a ) in lung tissues of rats with radioactive lung injury. Methods 72 female Wistar rats were randomly divided into the 3 groups, black control group (group A, n = 24 ), simple irradiation group( group B, n = 24 ), irradiation with TCM treatment group( group C, n = 24 ). The rats in group A and group B were given distilled water 2ml/200g by gavage for one week, however, the rats in group C were given TCM 2ml/200g(1.6g/200g) per day for one week, then the rats received irradiation,during irradiation,the rats' body weight was measured once a week, and the rats in group A and group B were given distilled water, and the rats in group C were given TCM by gavage till the end of the experiment. The expressions of TGF-β and TNF-a in lung tissue were detected at the fifth week( after lO-time irradiation)and at 12 weeks and at 26 weeks after irradiation. Results The expression of TGF-β protein in group B reached peak at the fifth week after irradiation ,which at different time points in group A and group C was significantly lower than that in group B ( P 〈 0.01 ). The expression of TNF-a in group B reached peak at the fifth week after irradiation, however, which in group C was obviously inhibited at the fifth week, and the expression of TNF-a in group C at the 5th week, 12th week,26th week was significantly lower than that in group B ( P 〈 0. 01 ). Conclusion Maimendong decoction has better preventive effect on radioactive lung injury, and its action mechanism is related with the inhibition of expression of TGF-βand TNF-a in lung tissues.
