摘要 为建立快速、灵敏的血清腺苷脱氨酶(ADA)匀相速率法检测新技术,通过对酶催化反应动力学体系、表面活性剂、液体酶稳定条件等关键技术研究,建立了嘌呤核苷磷酸化酶-黄嘌呤氧化酶-过氧化物酶(PNP-XT0-POD)偶联单一试剂可见光匀相速率法测定血清ADA体系。结果表明,该法的线性范围可达0.08~129.00 u/L,平均回收率为99.4%,批内变异系数和批间变异系数(CV)分别为0.013~0.031和0.022~0.041,与美国Diazyme Laboratores提供的ADA检测试剂方法比较具有良好的相关性,线性回归方程和相关系数分别为y=0.999 9 x+0.054 5,r=0.997 3。该法简便、快速,灵敏可靠,适合临床推广应用。 To establish a simple, sensitive method of single reagent spectrophotometric kinetic method of serum ADA analysis, by the right of choosing dynamics system of enzymic catalytic reaction, surfactants, conditions for stability in liquid enzymes, establishing PNP- XTO -POD -coupled single reagent spectrophotometric kinetic method of serum ADA analysis. The results show that the PNP - XTO - POD -coupled single reagent spectrophotometrie kinetic method of serum ADA analysis is; liner to 0. 08 129.00 u/L, the average rate of recovery is 99.40%, the withinrun and betweenrun CV is 0. 013 ~ 0. 031 and 0. 022 -0. 041. Comparing this methods with diazyme laboratores's ADA assay, the regression equations was obtained: y = 0. 999 9 x + 0. 054 5, r = 0. 997 3. It concludes this method is simple, sensitive and suitable for clinical application.
