摘要
本文采用碱性成纤维细胞生长因子(bFGF)和白介素一1(IL-1)所诱导的大鼠血管平滑肌细胞(VSMC)增殖模型.以MTT法观察了海洋硫酸多糖DPS对VSMC增殖的影响。大鼠血管平滑肌细胞在DPS浓度为0.001μg/ml~100μg/ml的培养液中孵育24h后,分别加入bFGF(50ng/ml)或IL-1(50U/ML)再孵育24h,用MTT法测定海洋硫酸多糖DPS村VSMC增殖的影响。结果表明,DPS在0.01μg/ml,0.1μg/ml,1μg/ml,10μg/ml浓度下,对bFGF或IL-1所致的VSMC增殖均有明显的抑制作用(P<0.01),最佳抑制浓度为1μg/ml,且DPS对VSMC增殖的抑制作用随着bFGF或IL-1浓度的增加而减弱。
The effects of D-polymannuronic sulfate (DPS)on the proliferation of rat vascular smooth muscle cells (VSMC)induced by basic fibroblast growth factor (bFGF)or interleukin-1 (IL-1 )were studied in this paper. Pretreatment with DPS in concentrations ranging from 0.001μg/ml up to 100μg/ml to rat aortic smooth muscle cells incubated at 37℃ for 24 hours,followed by addition of bFGF (50ng/ml )or IL-1 (50U/ml )for another 24 hours, respectively. The effects of DPS on the proliferation of VSMC was determined by MTT assays. The results indicated that anti-roliferative effects of DPS were observed at concentrations of 0.01 μg/ml, 0.01μg/ml, 0. 1μg/ml, 1μg/ml, 10μg/ml, with its maximal effect observed at concentration of 1μg/ml.Moreover, the suppressing actions of DPS on the proliferation of VSMC were diminished by increasing the concentrations of bFGF or I1-1,implicating that DPS is capable of producing an inhibitory effect on rat aortic smooth muscle cells proliferation induced by bFGF or IL-1.
