摘要
用噻唑蓝染色法测定屏边三七花粉不同保存条件下的活力和寿命,用联苯胺-过氧化氢法测定柱头可授性。结果表明,屏边三七花粉离开母体后活力较高,达到89.91%。在冷藏条件(4℃)下,贮藏时间在12d之内,屏边三七花粉活力的衰减程度小于20%,具有较高的活力;屏边三七花粉保存60d后完全失去活力;在冰冻条件(-20℃)下,屏边三七花粉保存35d后活力下降至2.05%,至46d后完全失去活力。屏边三七的柱头在小花开放后第8天开始具有强可授性,可授性高峰期持续时间为5d,柱头可授性在1d之中的高峰期为11∶00~16∶00。研究结果为杂交育种时间的选择提供依据。
The pollen viability of Panax stipuleanatus was detemained by M'IT,and the stigma receptivity was visualized by using benzidine- H202 reaction. The results showed that the pollen viability of Panax stipuleanatus was higher than 89.91% after leaving maternal. When pollen was preserved at 4℃ for 60 days,the viability was lost completely and at - 20℃ for 35 days,the viability attenuated to 2.05% and for 46 days the viability was lost completely. The stigmas of Panax stipuleantus had the highest receptivity for five days from the eighth day after flowering, the peak stigna receptivity stages were 11:00~ 16:00.The study pollen viability and stigma receptivity of Panax stipuleanatus for cultivar breeding by artificial hybridization.
