正交试验设计比较金银花药材中绿原酸与木犀草苷的水提工艺 被引量:10

摘要 目的通过正交试验比较金银花药材中绿原酸与木犀草苷的水提工艺。方法建立了HPLC法同时测定金银花提取物样品中绿原酸和木犀草苷含量的方法。分别采用煎煮和超声2种水提工艺提取金银花药材中的绿原酸和木犀草苷,正交试验设计安排实验,分别以绿原酸转移率、木犀草苷转移率和总转移率评分3种指标对正交试验结果进行分析与比较。结果以绿原酸转移率为指标,煎煮法优于超声法;以木犀草苷转移率为指标,则超声法优于煎煮法。同一提取方法,评价指标不同所得到的最佳工艺也不同,对2种成分的提取效果也有差异。结论在金银花的提取工艺研究中,不能仅以单一成分的转移率为指标进行评价,否则不能反映2种甚至多种成分的综合提取效果。 OBJECTIVE To compare the water extraction process of Chlorogenic acid and Luteolin-7-O-glucoside in Flos Lonicerae by orthogonal design.METHODS HPLC method was established to determine the content of Chlorogenic acid and Luteolin-7-O-glucoside in Flos Lonicerae extraction samples.Chlorogenic acid and Luteolin-7-O-glucoside in Flos Lonicerae were extracted by water decoction and ultrasound method using orthogonal design tests,and single extraction rate and total extraction rate of them were adopted as indexes to analyze and compare the results of orthogonal design tests.RESULTS Decoction method was better than ultrasound method according to extraction rate of Chlorogenic acid,and an opposite result was drawn according to extraction rate of Luteolin-7-O-glucoside.Different optimized process and extraction effect were obtained with different valuation indexes if extracted by identical method.CONCLUSION Extraction rate of single ingredient will not be recommended as evaluation index in investigating the extraction process of Flos Lonicerae,which could not reflect a reasonable extraction effect of two or more ingredients.
